Synthesis and Biological Evaluation of a Copper(II) Complex Incorporating a Multidentate Bipyridine-Carboxamide Ligand

Abstract

A novel Cu(II) complex containing a bipyridine-based amide ligand was successfully synthesized and characterized using FT-IR,H-NMR,13C-NMR, and mass spectrometry. Spectroscopic analyses confirmed the proposed chemical structures and indicated coordination to the Cu(II) center through nitrogen donor atoms. The antibacterial activities of both the free ligand and its Cu(II) complex were evaluated against Staphylococcus aureus and Escherichia coli. While the free ligand showed negligible antibacterial activity, the synthesized Cu(II) complex exhibited significant, concentration-dependent inhibitory effects against both bacterial strains. To complement the experimental findings, molecular docking studies were conducted against bacterial target proteins (PDB IDs: 6MH4 and 6QF6). The computational results revealed favorable binding energies of -6.49 and -6.46, kcal/mol, respectively, establishing stable non-covalent interactions within the active sites. These combined experimental and theoretical results demonstrate the promising antibacterial potential of the synthesized Cu(II) complex, highlighting its suitability for further bioinorganic and medicinal chemistry applications.
